The production of pancreatin usually adopts two methods: extraction, drying, and then defatting or the pancreas is dried, defatted, and then extracted.

Place the fresh porcine pancreas below room temperature with 20% dilute ethanol, add concentrated ethanol for precipitation, collect the precipitate, make a dry powder at low temperature (<5°C), and defatted it with ether.

Take 100 kg of the minced frozen pancreas and thaw at room temperature, put 25%-30% ethanol in the pot in advance (must be prepared a few days ago, the temperature should be below 10%) 15 kg, stir constantly, mix well, and keep it at room temperature. Then, 0~10℃, autolytic digestion for more than 48h, filter out pancreatic milk, pancreatic dregs, and then add the same amount of 25%-30% ethanol to extract once, the operation method is the same above, and the second pancreatic milk is combined.

Add 85% ethanol to pancreatic milk at 0-10°C to make the ethanol concentration reach 60%-70%, and mix well. Cool to 0～5℃ and let stand for 24h. If necessary, let it stand at -5°C to make the precipitation complete.

Siphon out the supernatant, and the lower sediment is pancreatin.

Put the precipitate in a cloth bag for hanging filtration, try to remove most of the ethanol, and then squeeze it to obtain a crude product.

The crude pancreatin is circulated, degreasing with ether until the fat is removed (paper trace test: take the ether extract and drop it on the filter paper. After all the ether is evaporated, there should be no fatty oil stains). Next, the degreasing pancreatin is dried in circulating hot air below 40°C and ground into a fine 60-80 mesh powder. After passing the sample test, we get pancreatin product.
